ATCC highly recommends that appropriate personal protective equipment is always used when handling vials. For cultures that require storage in liquid nitrogen, it is important to note that some vials may leak when submersed in liquid nitrogen and will slowly fill with liquid nitrogen. Upon thawing, the conversion of the liquid nitrogen back to its gas phase may result in the vial exploding or blowing off its cap with dangerous force creating flying debris. Unless necessary, ATCC recommends that these cultures be stored in the vapor phase of liquid nitrogen rather than submersed in liquid nitrogen.

1. Open vial according to enclosed instructions.
2. Using a single tube of broth, aseptically transfer 0.5 ml to the vial and rehydrate the pellet.
3. Aseptically transfer this aliquot back to the broth tube and mix well. Transfer 0.1 ml to slants. Plate the rehydrated culture (0.1 ml ) onto non-selective medium (to test for purity).
4. Incubate the culture at 30oC for 72 to 96 hours. When using ATCC Medium #1306 the culture needs to be fed a mixture of 50% methane and 50% air every 24 hours for best results.
5. Growth should be detected within 4 to 6 days. No growth should be detected on the non-selective plates.

Microscopic examination shows the cells to be coccoid-shaped, motile, and tend to grow in clusters. Cells will grow on agar.
